Background
Myf5 (Myogenic Factor 5) is a basic helix-loop-helix (bHLH) transcription factor encoded by the MYF5 gene and a key member of the myogenic regulatory factor (MRF) family, essential for skeletal muscle development. Structurally, it contains a DNA-binding domain that recognizes E-box sequences in muscle-specific gene promoters. Myf5 is expressed in the dermomyotome of early somites, where it commits myogenic precursor cells to muscle differentiation, as well as in brown adipose precursors and the neural tube during embryonic development. In adults, it serves as a marker of satellite cells, the muscle stem cells responsible for regeneration. Functionally, Myf5 acts as a transcriptional activator, initiating muscle gene expression and coordinating myogenesis alongside other MRFs like MyoD and myogenin. Understanding Myf5's role in muscle formation has therapeutic implications in muscle regeneration, muscular dystrophies, and regenerative medicine.
