Background
Wnt16 is a crucial regulator of skeletal homeostasis, playing a vital role in maintaining cortical and trabecular bone integrity while modulating osteoarthritis (OA) progression. It balances bone remodelling by influencing osteoblastogenesis and osteoclastogenesis through canonical and non-canonical Wnt signalling pathways mediated by receptors such as Ror2, CD146, and LRP6. Wnt16 suppresses osteoblast maturation and mineralization but enhances bone mass by regulating transcription factors like TBX5, TBX15, USF3, and TWIST1/TCF12, promoting osteoblast differentiation and inhibiting osteoclastogenesis. In OA, Wnt16 protects cartilage by inhibiting IL-1β-induced catabolic factors (e.g., MMP13, ADAMTS5), preventing chondrocyte hypertrophy, and maintaining cartilage anabolism through lubricin upregulation. Wnt16 prevents the overactivation of canonical Wnt signalling, which can worsen OA, while also interacting with pathways such as mTOR and JNK to maintain cartilage homeostasis and protect against degeneration.
