PSMD11 is a critical subunit of the 19S regulatory particle (RP) within the 26S proteasome, playing a pivotal role in the ubiquitin-proteasome system by facilitating the recognition, unfolding, and deubiquitination of damaged or misfolded proteins for degradation. It is essential for maintaining protein homeostasis, regulating cellular processes such as signal transduction, and combating oxidative stress, which is particularly important in aging and age-related diseases like neurodegeneration and presbycusis. PSMD11 interacts with AMPKα2, a cellular energy sensor, to enhance its protective functions under stress conditions. Mutations in PSMD11 can lead to proteasomal dysfunction, resulting in neurodevelopmental disorders characterized by cognitive delays, speech and motor impairments, and metabolic issues like early-onset obesity. Additionally, PSMD11 dysfunction triggers type I interferon activation, linking it to immune dysregulation.
